Real-time fluorescence quantification PCR technology refers to the addition of fluorescent dyes or fluorescent groups into the PCR reaction system, the use of fluorescence signals to monitor the whole PCR process in real time, and finally through the standard curve for quantitative analysis of unknown template concentration.
Its characteristics are as follows:
(1) In order to avoid the inaccuracy of the end-point quantification, real-time dynamic and continuous fluorescence monitoring is carried out by using the indicator which produces fluorescence signal to display the amount of amplification products, and the pollution of samples and products is eliminated, and there is no complex follow-up processing of products.
(2) The fluorescence signal is obtained by embedding fluorescent dye into double stranded DNA, or by combining fluorescent probe with wood extract, which improves the sensitivity, specificity and accuracy of detection. Real time o-pcr can be applied to the study of mRNA expression, the detection of DNA copy number, the detection of single nucleotide polymorphism, the analysis of cytokine expression, the study of tumor resistance gene expression and the quantitative monitoring of virus infection.
